Few cellular models of VEXAS syndrome have been developed as of the date provided. An inducible human iPSC model has been described, but the induction of UBA1 mutations leads to a rapid loss of mutant clones. PKAcα Mutation and PKA Inhibition in Fibrous Dysplasia Mouse Model

Researchers generated a constitutively active mutant of PKAcα by introducing a W197R amino acid substitution via site-directed mutagenesis. Compared to the wild-type, this mutant exhibited increased transcriptional activity of the cAMP-responsive element luciferase reporter in 293T cells. FD lesions were confirmed by histopathological examinations revealing features such as a fibrous matrix replacing normal lamellar bone, abundant osteoclasts suggesting bone resorption, and mineralization defects. Endogenous protein kinase inhibitor peptide PKI was used to inhibit PKA activity effectively, suppressing phosphorylation induced by forskolin and 3-isobutyl-1-methylxanthine. Each group reported one severe hypoglycemia incident, while diabetic ketoacidosis occurred in two participants in the closed-loop group and one in the standard care group. Denice Feig, MD, noted that participants found it easy to learn to use the hybrid closed-loop system during pregnancy. Manchester City was defeated 4-3 by Saudi Arabian side Al Hilal in the last 16 of the Club World Cup on June 30 at the Camping World Stadium.
