According to Bank of England figures, around 63,000 mortgages were approved for house purchase in May, representing an increase of approximately 2,400 from April. This was the first increase in approvals since December 2024. Approvals for remortgaging with a different lender also rose by 6,200 to 41,500 in May, marking the largest increase since February 2024. Stamp duty discounts in England and Northern Ireland became less generous from April. Recent changes in mortgage affordability assessments have been made by several lenders following clarification from the Financial Conduct Authority (FCA). Small and medium-sized non-financial businesses borrowed a net £0.4 billion, following flat net borrowing in April. All mice in the study were kept on a C57BL/6J genetic background and housed under a regulated environment at the University of Texas Southwestern Medical Center (UTSW). Stem cell research was conducted under UT Southwestern Stem Cell Research Oversight Committee's approval (registration no. 8). HEK293T cells and a human iPS cell line (WTC-11, GM25256) were used, both of which were cultured according to specified protocols. Rat primary cortical astrocytes were also cultured according to Invitrogen protocols. The SW102 cell line was obtained from the National Cancer Institute. A BAC containing the full-length human CLOCK gene was acquired from BACPAC Resources. The cesium chloride-purified human CLOCK BAC was integrated into C57BL/6N mice by direct pronuclear injection. Founders were screened by RT-qPCR, identifying three founders expressing the FLAG-tagged human CLOCK. Mice from these lines were backcrossed onto a C57BL/6J genetic background for ten generations, with line HUf7 being used for all experiments due to its higher CLOCK expression. Genomic DNA was extracted from mouse ear or tail snips and used for PCR and RT-qPCR analyses to detect human CLOCK and mouse Clock expression. Mice were subjected to a variety of tests, including behavioral analysis, physiological measurements, and molecular analyses to study the effects of CLOCK expression. Study on Monkeypox Virus Infection in Human Neural Organoids from Stem Cells

Human neural organoids (hNOs) were generated from human pluripotent stem cells, specifically H1 embryonic stem cells and IMR90 induced pluripotent stem cells, according to established methods. The cells were observed and characterized using immunofluorescence markers to distinguish between neurons and neural progenitor cells (NPCs) over a 112-day developmental period. During development, the hNOs exhibited a structure that included small ventricular units and TUJ1-positive neurons. By the second month, the organoids developed a layered organization with enlarged ventricles surrounded by NPCs and a population of SOX2-positive cells within a neuron-dominated environment. Astrocytes appeared by the third month of maturation. Eight batches of H1-derived hNOs and three batches of IMR90-derived organoids, cultured for 70-110 days, were exposed to a 2022 primary isolate of the monkeypox virus (MPXV) clade IIb. SOX2-expressing NPCs, neurons, and GFAP-expressing astrocytes showed susceptibility to MPXV infection, with viral antigen localizing to these cells. To corroborate findings, 2D cultures enriched for NPCs and forebrain neurons were also subjected to MPXV infection, confirming both NPCs and forebrain neurons harbor viral antigen. A low multiplicity of infection was applied to assess viral replication dynamics over 10-14 days. Immunofluorescence analysis revealed increasing viral antigen presence over time, with maximal intensity observed between days 10 and 14. MPXV tended to cluster in foci at the surface layers of hNOs. Tests were also conducted on 2D cultures and confirmed the presence of viral antigen within cell extensions. Transmission electron microscopy identified pleomorphic intracellular mature virions in infected cells. The vaccinia-related MPXV virus trafficking involved neurites and possibly through neuron-to-neuron transmission. Titrations revealed that hNOs predominantly retained MPXV within cell-associated compartments compared to released virus. Forebrain neurons revealed preferential cell-associated transport dynamics, delaying the reproduction of extracellular infectious virus.
